(1) Formula
① 200g potato, 20g glucose, 5g peptone, 18-20g agar, 3g potassium dihydrogen phosphate, 1.5g magnesium sulfate, pH 6.5-7.0, 1000mL water.

② 200g potato, 20g glucose, 50g wheat bran, 18-20g agar, 3g potassium dihydrogen phosphate, 1.5g magnesium sulfate, pH 6.5-7.0, 1000mL water.

(2) Inoculation and Cultivation
Prepared, sterilized, and inoculated according to standard procedures, incubated at 25℃ in the dark.

(3) Labeling and Cultivation: Before removing the test tubes from the inoculation box, label each tube with the strain number and inoculation date. Then, place the tubes in an incubator (room) at 23–25°C for 8–10 days until the culture slant is fully grown.

(4) Strain Preservation: Select uncontaminated culture slants, wrap them in paper, and store them in a refrigerator at 4°C.
