① The culture room must be dry, clean, dim and well ventilated, and fitted with insect prevention facilities. Thoroughly clean and disinfect the room before use. Use aerosol disinfectant at 4–6 grams per cubic meter, seal the room tightly and fumigate for 24 hours. Regular disinfection is also needed throughout the whole mycelium cultivation period.
② Most ordinary mushroom cultivation sheds have no heating equipment. Low-temperature culture will not seriously affect mycelium growth, and can effectively reduce miscellaneous bacteria contamination, but it will extend the spawn run period and delay fruiting time. Properly raising indoor temperature can shorten mycelium growing cycle and ripening period. The ideal growth temperature for shiitake mycelium is 20–25°C.
③ Shiitake mushroom is aerobic. Insufficient oxygen will slow down or stop mycelium growth. Timely ventilation is essential to exhaust harmful gas, reduce bacteria concentration and boost mycelium activity. If you feel stuffy or smell strong substrate odor when entering the room, ventilation must be carried out immediately.
④ After hole punching for aeration, oxygen inside spawn bags increases sharply and speeds up mycelium metabolism. The internal bag temperature is usually 3–5°C higher than ambient temperature. Long-term temperature above 30°C together with oxygen shortage will cause mycelium weakness, shrinkage and even death. It is vital to prevent high temperature burn in later cultivation stage.
Start temperature monitoring 20 days after inoculation. Once the stack temperature exceeds 28°C, open windows for ventilation or reduce stacking quantity to control bag temperature below 28°C. Keep away from strong sunlight and rain. Maintain indoor relative humidity below 60% to effectively prevent miscellaneous bacteria infection.
Special Reminder: Four Taboos in Shiitake Mycelium Cultivation
- Avoid spawn bag damage. Even tiny cracks will lead to bacterial infection easily.
- Avoid rain soaking. Wet spawn bags are highly prone to contamination after inoculation.
- Avoid direct sunlight. Strong UV rays damage mycelium, cause material fermentation and loose substrate.
- Avoid violent shaking and dropping. Severe vibration will induce premature budding, hinder vegetative growth and delay normal maturation. Please handle spawn bags gently in all operations.
