Inoculation with liquid strains
1. Timely inoculation
The liquid strains should be connected to the mushroom culture medium when they are in the most active state, and the best applicable period is only one or two days. If they are overdue, problems such as aging of the age of the bacteria and autolysis of the bacteria balls will occur. If they are used reluctantly, the yield and quality of the finished mushrooms will be affected. Therefore, special attention should be paid to the connection arrangement of production.
2. Fine operation
Fine operation is closely related to the yield of the mushroom bottle (bag). At present, large production enterprises or factory-based bottle cultivation generally use efficient and automated inoculation equipment, while small enterprises and factory-based bag cultivation mostly use inoculation guns for manual inoculation. Although there are differences in efficiency between the two, the standard requirements should be basically the same, that is, the liquid volume should be accurate, the spraying should be uniform, and the coverage should be complete.
3. Inoculation with efficient and automated inoculation equipment
Push the fermentation tank directly next to the inoculator, and place the inoculator under a large purification hood. During inoculation, the flow of personnel and goods will not affect the environment of the inoculation area, and the inoculation effect is very stable. The inoculator completes the uncovering, digging, inoculation, and capping.
a. Carry out a pre-startup inspection; if all parts are normal, disinfect the inoculator and nozzle, connect the fermentation tank, and set the inoculation volume; each bottle receives 30-35ML of liquid culture.
b. Before inoculation, the cooling room staff is responsible for checking the temperature of the cultivation bottle material. The material temperature should be less than 20℃ before inoculation.
c. Start the machine and start inoculation.
d. Before or during inoculation, if the inoculator or other related equipment fails, it should be reported in time. After the mechanical repair personnel of the Engineering and Equipment Department repair it, the inoculator is disinfected and inoculation begins, and the abnormal reason is written on the "Inoculation Room Record Sheet".
e. The entire inoculation process is strictly in accordance with the requirements of the "Clean Area SOP".
f. During the inoculation of each tank, the inoculator is responsible for self-inspection of the inoculation volume, and each tank is measured once before and after inoculation. The average inoculation volume of each tank is required to be 30~35ml. If the deviation is large, the machine should be stopped and readjusted, and the relevant important parts that have been touched by people should be disinfected.
